Tyrosinase@HKUST-1: a super stable biocatalyst efficient for catecholic product synthesis

Abstract Although metal–organic frameworks (MOFs) have been considered as promising matrices for enzyme immobilization, HKUST-1, constructed from copper acetate (CuAc 2 ) and benzene 1,3,5-tricarboxylate (BTC), has rarely explored this application. In study, mushroom tyrosinase (EC 1.14.18.1) was immobilized in the form of tyrosinase@HKUST-1 following a simple reaction procedure by mixing BTC with prior to addition CuAc . The resultant biocatalyst characterized both structural features catalytic properties. Upon incorporation into HKUST-1 frameworks, gained prominent enhancement stability against pH, temperature storage: When incubated at 50 °C pH 6.0, presented half-life 32.6 h, which is 77-fold over tenfold higher than that free its other immobilization forms, respectively; catalyst fully maintained activity least months when stored 30 °C. applicability new demonstrated employing it regioselective ortho -hydroxylation reactions produce catecholic products huge pharmacological effects, i.e., hydroxytyrosol l -DOPA, excellent yields productivities.
